Interactions between proteins play a significant role in biological processes, including those involved in disease. The team behind the SIMPL2 platform designed it to optimize researchers' ability to measure protein-protein interactions for targeted drug therapies. While protein-protein interactions have previously been considered 'undruggable' using small molecules , the platform addresses this challenge by facilitating the measurement of these interactions—improving our understanding of the types of molecules needed to control them. "Many methods have been developed to measure interactions between proteins, especially more recently as the significance of protein interactions in disease has become more apparent," said Zhong Yao, first author on the study and senior research associate of U of T's Donnelly Centre for Cellular and Biomolecular Research. "However, all of these methods have shortcomings, including high costs and complicated procedures that delay results. The biggest advantages of our SIMPL2 platform are that it produces more reliable measurements and is comparatively cheaper to use." The study was published recently in the journal Molecular Systems Biology . Yao started working on the protein interaction measurement problem while developing the original SIMPL (Split-Intein Medicated Protein Ligation) system . SIMPL2 is an update of SIMPL that involves the use of the split luciferase enzyme for detection of protein interactions through luminescence. In addition to improving identification of interactions, the entire measurement process occurs through one medium: liquid. This simplifies the process considerably by reducing the number of steps required to carry out measurements. "One of the issues with SIMPL was that we had to use an additional process, called ELISA, to identify the proteins spliced by the SIMPL platform," said Yao. Moylan on Thursday sent a letter to Gov. Lou Leon Guerrero requesting a meeting to reprogram $100 million ARP funding to GHURA, especially considering GHURA’s apparent violation of the Open Government Law that makes all their actions void and voidable. The AG said there remains only five days left before the deadline to legally program the $100 million that the community “desperately needs for our people’s daily health care, education and public safety problems.” Moylan said the Guam Memorial Hospital complains about a failing hospital threatening health care, the school board complains about school facilities threatening kids’ safety and education and Department of Corrections complains about an overcrowded prison. The AG said he is available to meet 24/7 and suggested to invite elected senators to consult on how best to program the $100 million which must be legally spent under Guam law by Dec. 31. “They may be helpful to advise you about their views in assisting to commit additional local funding for our people’s welfare,” Moylan wrote to the governor. With that deadline approaching, there was no movement in GHURA’s condemnation case over 42 acres of land for the hospital beyond the initial complaint and notice filed in the Superior Court of Guam as of Thursday. Moylan filed a complaint for declaratory and injunctive relief in federal court after the governor and GHURA both disagreed with his legal opinions that they violated the law. Previously, Moylan issued legal opinions that found GHURA violated the law when it accepted $12.5 million to pay for the land from the governor, and when it moved forward with condemnation proceedings. The AG’s office is asking the court for a writ of mandamus and request for injunctive stay to prohibit the governor and GHURA from spending any additional American Recovery Plan Act funding. The AG’s office is also asking the court to void any actions taken by GHURA for the Mangilao medical complex including invalidating the loans it got from the governor. It is also asking the court to prohibit GHURA from using or transferring the land it has purchased to date until authorized by the Legislature or unless consistent with its purposes. The governor has allocated $100 million in ARP money to install infrastructure at the Mangilao hospital site. The suit alleges the governor “will continue to unlawfully spend funds,” in violation of the Organic Act of Guam, unless the court orders her not to use ARP money for the purpose of obtaining land without legislative authorization. Adelup said the American Rescue Plan Act was passed in 2021 and the AG has been in office for almost two years. “If he truly believed these funds needed to be reappropriated before they were utilized, he could have and should have brought his suit earlier, instead of waiting for the eleventh hour to complain only about a single project that he happens to disagree with,” Adelup said. The governor’s office also argued that the law in this area is well-settled. The Legislature has no control over federal grant funds that are given for a specific purpose, are fully federal and have no requirement to be under local control. The AG has said $100 million could be used to immediately fix the Guam Memorial Hospital versus spending $1 billion for a medical complex that will take a decade to complete and has no funding to finish. Adelup said because the AG is seeking an injunction, court rules require him to provide a bond in an appropriate amount to pay damages if he is wrong. “Here, that could mean Guam loses out on over $100 million in federal funds. If the AG insists on filing baseless complaints, he better be prepared to pay for it. Seery, of Vilanstown, Gaybrook, Mullingar, Co Westmeath, was also ordered to carry out 240 hours of community service instead of a sentence of two years and six months for assault causing harm to Garda Gemma Prendergast. He must also pay each victim €5,000. Judge Johnson recalled the prosecution evidence, which revealed how Seery used an Instagram account with an AI-generated name. He began communicating with the girl, “persistently” asking for nude pictures. He told her he would send her an iPhone 12 and she sent him eight to 10 photos, including two without clothes, but edited them so her face was not visible. The phone did not arrive, and when queried, “the accused threatened to send the pictures to the victim’s followers if she did not meet and have sex with him”. It was then arranged that they would meet at a bridge near the old abandoned train station in Mullingar, but the victim reported him to gardaí. Gda Prendergast went to the area and presented herself as the victim, but Seery realised “and reacted by punching her”. The officer identified herself as a garda, and “the accused proceeded to kick her in the head”. She described the blows as “exceptionally violent”. She managed to get her hand up to protect her head from the force of the kick, which sent her “spinning to the ground” and leaving her bruised. Other gardaí observing the incident arrested the accused, who admitted he was behind the Instagram account. The girl’s victim impact statement outlined the extreme upset caused, and she told gardaí “she wants to put the matter behind her and move on with her life”. In his interviews, Seery told gardaí he had no friends besides people he engaged with online. Gda Prendergast said in her victim impact statement that, in her 20-year career, she had never encountered such aggression before and it has left her shaken. Judge Johnson noted: “Instead of trying to talk or run away, his first response was to kick her straight in the head.” Judge Johnson described the offences as disgraceful and cowardly, involving a vile intrusion on the girl. Seery then adopted a sinister approach and attempted to blackmail her into having sex with her, or the pictures would be distributed to her followers. “This was an appalling and despicable act and clearly involved creative planning and premeditation,” he said, adding that this was a particularly aggravating feature of the case. Both victims were traumatised, he noted. A forensic psychological assessment put found Seery’s risk of further sexual offence in the medium range, based on his current status, but found it could increase if he began drinking heavily again. Mitigating factors were his young age, remorse, lack of previous convictions, and guilty plea, and the judge said it appeared Seery’s offending was affected by his sense of isolation and depressive symptoms. He said it was out of character for the accused and an aberration. It was clear, he stated, that Seery was from a good family, and following apprehension, he accepted the offences were malicious, furnished letters of apology to his victim, and expressed shame. The court heard he worked part-time, was on a film production course, and engaging with his doctor and counselling, which needs to continue, and he has quit binge drinking. The court noted he had been recently diagnosed with autism, and this diagnosis would assist him in getting help to reduce his risk of reoffending. The judge learned from the accused’s father that Seery had issues from childhood, suffered a mental breakdown in his teens, and “was in a dark place for years”. The accused, who had no prior convictions, will be placed on the sex offenders register, must not reoffend for six years, must never contact the victims, continue counselling, and remain in education and probation supervision for 18 months.

JERUSALEM — A new round of Israeli airstrikes in Yemen on Thursday targeted the Houthi rebel-held capital and multiple ports, while the World Health Organization's director-general said the bombardment occurred nearby as he prepared to board a flight in Sanaa, with a crew member injured. "The air traffic control tower, the departure lounge — just a few meters from where we were — and the runway were damaged," Tedros Adhanom Ghebreyesus said on social media. He added that he and U.N. colleagues were safe. "We will need to wait for the damage to the airport to be repaired before we can leave," he said, without mentioning the source of the bombardment. U.N. spokesperson Stephanie Tremblay later said the injured person was with the U.N. Humanitarian Air Service. Israel's army later told The Associated Press it wasn't aware that the WHO chief or delegation were at the location in Yemen. The Israeli military said in a statement it attacked infrastructure used by the Iran-backed Houthis at the international airport in Sanaa and ports in Hodeida, Al-Salif and Ras Qantib, along with power stations, claiming they were used to smuggle in Iranian weapons and for the entry of senior Iranian officials. Israel's military added it had "capabilities to strike very far from Israel's territory — precisely, powerfully, and repetitively." The strikes, carried out more than 1,000 miles from Jerusalem, came a day after Israeli Prime Minister Benjamin Netanyahu said "the Houthis, too, will learn what Hamas and Hezbollah and Assad's regime and others learned" as his military has battled those more powerful proxies of Iran. The Houthi-controlled satellite channel al-Masirah reported multiple deaths and showed broken windows, collapsed ceilings and a bloodstained floor and vehicle. Iran's foreign ministry condemned the strikes. The U.S. military also targeted the Houthis in recent days. The U.N. says the targeted ports are important entryways for humanitarian aid for Yemen, the poorest Arab nation that plunged into a civil war in 2014. Over the weekend, 16 people were wounded when a Houthi missile hit a playground in the Israeli city of Tel Aviv, while other missiles and drones were shot down. Last week, Israeli jets struck Sanaa and Hodeida, killing nine people, calling it a response to previous Houthi attacks. The Houthis also have been targeting shipping on the Red Sea corridor, calling it solidarity with Palestinians in Gaza. The U.N. Relatives and friends mourn over the bodies of five Palestinian journalists Thursday who were killed by an Israeli airstrike in Gaza City at the Al-Aqsa Hospital in Deir al-Balah. Meanwhile, an Israeli strike killed five Palestinian journalists outside a hospital in Gaza overnight, the territory's Health Ministry said. The strike hit a car outside Al-Awda Hospital in the built-up Nuseirat refugee camp in central Gaza. The journalists worked for local news outlet Al-Quds Today, a television channel affiliated with the Islamic Jihad militant group. Islamic Jihad is a smaller and more extreme ally of Hamas and took part in the Oct. 7, 2023, attack in southern Israel that ignited the war. Israel's military identified four of the men as combat propagandists and said that intelligence, including a list of Islamic Jihad operatives found by soldiers in Gaza, confirmed that all five were affiliated with the group. Mourners cry Thursday while they take the last look at the body of a relative, one of eight Palestinians killed, during their funeral in the West Bank city of Tulkarem. Separately, Israel's military said a 35-year-old reserve soldier was killed during fighting in central Gaza. A total of 389 soldiers have been killed in Gaza since the start of the ground operation. The war began when Hamas-led militants stormed across the border, killing around 1,200 people, mostly civilians, and abducting about 250. About 100 hostages are still inside Gaza, at least a third of whom are believed to be dead. Israel's air and ground offensive has killed more than 45,000 Palestinians, according to the Health Ministry. It says more than half the fatalities are women and children, but doesn't say how many of the dead were fighters. The offensive caused widespread destruction and hunger and drove around 90% of the population of 2.3 million from their homes. (NYSE:ATI)SINGAPORE: The sun is just peeking through on a Friday morning when I speak to Raymond Webster. It's also after midnight in the United Kingdom, where he lives, but Webster assures me his bedtime is late. After all, this isn’t your average 70-year-old. The Englishman is an avid mountain climber and passionate about driving. When not doing either of those things, he competes in the professional badminton circuit against others more than half his age. Since entering his first Badminton World Federation event in 2016, he's mostly hovered around the 800 mark in the men's rankings, and played close to 60 singles matches. He has lost all of them. Webster's recent matches include a 3-21, 8-21 defeat to 23-year-old Bruneian Kan Kah Kit in November; and a lopsided 3-21, 1-21 loss to 29-year-old compatriot Ngan Heng Lin in September. Webster loved every minute of these contests. To understand what drives him in his badminton exploits, one has to wind the clock back about five decades. "WILD EXPERIENCES" Webster’s foray into the sport began in his 20s, when he joined coaching sessions by top English player Ray Stevens. “Ray has a magnetic personality and fire for the sport that would set anyone's ambitions alight,” Webster told CNA. Though he was regularly "pummelled" by Stevens, badminton started to appeal to him for its "pure dynamics and the way the game flowed from surges of all-out intensity to moments of fine control”. It was during one of these sessions when "the lights came on" and Webster thought he could excel at the sport. "With a score of 11-15, it was the closest that I'd ever come to beating a champion," recalled the Englishman. "After the session, Ray asked me: 'How did you play like that?' Responding, I (said I) don't know. But years later I found out and fully understand that it stems from fight or flight. I have been in flow state several times on court." Webster next planned to head to China to play badminton, but this fell through. After scouring badminton magazines for contacts, he set off "with some trepidation" on his first trip abroad and travelled through the Americas for about four weeks. He still remembers his first event - a "dog fight" where he won with a third set score of 17-16 - as well as what it was like to play breathless at an altitude of 2,200m at the Mexican Open. Webster then spent five years in South Africa, working and playing provincial badminton. When he finally returned home, most of his former sparring partners had moved on to playing at the top level. Around this time, he picked up a recurring stomach injury and had to eventually forgo the sport he so loved. "Every time I moved, it was like I had glass inside me or wire mesh," he recalled. "I lost the ability to be active." At 42, Webster went back to university and eventually landed an engineering job in the semiconductor industry. It meant he could travel the world and pursue his other hobby of mountain climbing. "Despite carrying a lot of pain, I climbed solo most weekends for 11 years," he said, adding that he was at his fittest at 50 years old "with the cardio power of a husky dog". "Although the wild experiences I had climbing in winter were a tremendous high, I longed to play badminton again ... (but) I thought I would never be able to." STEPPING INTO A "SECOND LIFE" Then came the toughest years of Webster's life, which started when he was made redundant at his job. He tried to play badminton again but it was too painful to do so, due to his old injury that seemed like it had no way to heal. "There seemed no hope and I hit a brick wall and without the job and the mountains and with the gremlin of stress ... I was in a deep mental well, depressed beyond function," he said. "And that is a horrible place to be." "I had no way out but to dig my way up. It's all I could do basically. A ll I knew at that time was to just do something physical so I would get on my bike and ride till I was tired." At the same time, he continued trying ways to regain what he called his "competitive mobility", while battling through the pain of the injury. Eventually, what helped was yoga. "With persistence, I became more flexible than when I was a teenager," said Webster. He slowly took to the badminton courts at his local club and before he knew it, at age 62, he was playing at his first BWF event. "The world of badminton had changed in every aspect, and walking onto a show court was like a time warp," said Webster. "Looking back I had been playing in black-and-white TV, and now stepped into full-colour HD. It was simply awesome." In the years since, Webster has often been on the end of lopsided scorelines. "When I first entered the tour, I had aims of more success, but it's a strong tide to swim against when I never played as a junior," he said. "A couple of times I had to play with flu or migraine and suffer the consequences. But I accept it. "It's akin to climbing those big mountains again, when bad weather beat me back, and I went up again for a fifth attempt in total adversity, scaring myself to the point of my feet trembling me off the ice footholds that I cut, and pushing beyond where I could not reverse." Still, there have been memorable moments. There was the first time he won a game on the BWF circuit, when he beat the Netherlands' Rakesh Sharma 23-21 before losing 26-28, 15-21. It was 2022 and his opponent was 34 years old then. "Being out on a badminton court when my opponents are generally 40 to 50 years younger, people tell me that is a victory," said Webster. "When I have performed well and the umpire says: 'respect', I feel too that I am playing for them, since most of them are former players and they too must stay engaged with the 'addiction'. "There're so many people that wish they were on court playing because they either were badminton players or want to be a badminton player. But for many reasons they can't play ... I feel like that I'm fulfilling their desire." With his story garnering some attention online, fans have requested wefies and spare shirts while others ask how he's maintained his mobility to play a sport that can be tough on the body. "Much of that is because my whole family were incredibly active. I never stopped moving as a kid, I climbed trees, cycled thousands of miles a year, 200 miles in a day and went climbing most weekends," he said. "It made me tough and durable (and) my thought processes have not changed or aged in any way." LIKE OPENING A GIFT On the BWF tour, players accumulate points by playing in and winning matches. Those with more points automatically enter the main draw of tournaments. Players like Webster need to compete in the qualifiers to make it to the main draw. "For myself, I am not aiming for ranking, just to maintain enough points to stay on the ladder and climb sometimes," he said. Getting to each tournament is the culmination of "a lot of organising" for the Englishman. "Sometimes I've played three events in succession and loaded the car with three sets of gear, flown back to the airport, changed all the gear, and off on the next adventure. It's a real buzz." None of this comes cheap, though. Travelling the world to compete requires money - and Webster has been digging into his savings to finance his passion. "I ask for second meals on the plane so I don't have to buy a dinner in a restaurant. And I try and bum an entry into an airport lounge and eat well," he added. "But I'm running on the sheer effort that I put into my career, it paid well and just enough to have kept me on the circuit for a few years." There've even been times when he spent nights in his car or in the airport. But when he finally gets on court, it's like "waiting for a gift and then opening it", said Webster. That sheer joy keeps him going. "I accept the low scores and nobody's expecting me to win. But what is one of the greatest motivations - apart from that I enjoy being in that environment, and I enjoy my body able to rally and move pretty well - is (having) people encourage me," he said. "My family, just siblings now, love what I engage myself in, and since I picked myself up and battled my way back from rock bottom and years of pain, it has opened new doors. "It's my second life and I aim to never stop. Or I'll drop." It's not about just taking part either: Webster is adamant that he can improve. Next year, he plans to head to the mountains to build up his lung power, before competing in a series of tournaments. "I know I have the inherent skills, but not the consistency. That will come with the right enabler," he said. Soon after our interview, Webster tells me he's mulling over competing at the Korea Masters. Initially, it seems as if the 30-hour journey to Iksan city might be too much of a disincentive. But when I check the results a couple of days later, I spot a familiar name. Webster made it to Korea after all. “When people ask my age, I reply 'I stopped counting at 21',” he said. “If age enters my head, I throw it out. I am not going to slow down.”
